Representing species-specific proteins and protein complexes in ontologies that are both human and machine-readable facilitates the retrieval, analysis, and interpretation of genome-scale data sets. Although existing protin-centric informatics resources provide the biomedical research community with well-curated compendia of protein sequence and structure, these resources lack formal ontological representations of the relationships among the proteins themselves. The Protein Ontology (PRO) Consortium is filling this informatics resource gap by developing ontological representations and relationships among proteins and their variants and modified forms. Because (...) proteins are often functional only as members of stable protein complexes, the PRO Consortium, in collaboration with existing protein and pathway databases, has launched a new initiative to implement logical and consistent representation of protein complexes. We describe here how the PRO Consortium is meeting the challenge of representing species-specific protein complexes, how protein complex representation in PRO supports annotation of protein complexes and comparative biology, and how PRO is being integrated into existing community bioinformatics resources. The Protein Ontology provides terms for and supports annotation of species-specific protein complexes in an ontology framework that relates them both to their components and to species-independent families of complexes. Comprehensive curation of experimentally known forms and annotations thereof is expected to expose discrepancies, differences, and gaps in our knowledge. We have annotated the early events of innate immune signaling mediated by Toll-Like Receptor 3 and 4 complexes in human, mouse, and chicken. The resulting ontology and annotation data set has (...) allowed us to identify species-specific gaps in experimental data and possible functional differences between species, and to employ inferred structural and functional relationships to suggest plausible resolutions of these discrepancies and gaps. Der aktuelle Eurozentrismus führt dazu, dass Europa und hier insbesondere die Europäische Union verschiedene Entwicklungen und Ereignisse in den internationalen Beziehungen falsch auffassen und interpretieren. Der Autor setzt sich thesenhaft mit diesem Phänomen auseinander und zeigt auf, wie es durch eine eurozentristische Sichtweise, die er als Überbewertung europäischer Interessen, Bedürfnisse, Strukturen und Ereignisse versteht, zu einer verzerrten Wahrnehmung der internationalen Politik durch die Europäer kommt.

Ran is one of the most abundant and best conserved of the small GTP binding and hydrolyzing proteins of eukaryotes. It is located predominantly in cell nuclei. Ran is a member of the Ras family of GTPases, which includes the Ras and Ras‐like proteins that regulate cell growth and division, the Rho and Rac proteins that regulate cytoskeletal organization and the Rab proteins that regulate vesicular sorting. Ran differs most obviously from other members of the Ras family in both its (...) nuclear localization, and its lack of sites required for post‐translational lipid modification. Ran is, however, similar to other Ras family members in requiring a specific guanine nucleotide exchange factor (GEF) and a specific GTPase activating protein (GAP) as stimulators of overall GTPase activity. In this review, the multiple cellular functions of Ran are evaluated with respect to its known biochemistry and molecular interactions. (shrink)

The Protein Ontology (PRO) provides a formal, logically-based classification of specific protein classes including structured representations of protein isoforms, variants and modified forms. Initially focused on proteins found in human, mouse and Escherichia coli, PRO now includes representations of protein complexes. The PRO Consortium works in concert with the developers of other biomedical ontologies and protein knowledge bases to provide the ability to formally organize and integrate representations of precise protein forms so as to enhance accessibility to results of protein (...) research. The Protein Ontology (PRO) web resource provides an integrative framework for protein-centric exploration and enables specific and precise annotation of proteins and protein complexes based on PRO. Functionalities include: browsing, searching and retrieving, terms, displaying selected terms in OBO or OWL format, and supporting URIs. In addition, the PRO website offers multiple ways for the user to request, submit, or modify terms and/or annotation. We will demonstrate the use of these tools for protein research and annotation.

The Protein Ontology (PRO; http://proconsortium.org) formally defines protein entities and explicitly represents their major forms and interrelations. Protein entities represented in PRO corresponding to single amino acid chains are categorized by level of specificity into family, gene, sequence and modification metaclasses, and there is a separate metaclass for protein complexes. All metaclasses also have organism-specific derivatives. PRO complements established sequence databases such as UniProtKB, and interoperates with other biomedical and biological ontologies such as the Gene Ontology (GO). PRO relates to (...) UniProtKB in that PRO’s organism-specific classes of proteins encoded by a specific gene correspond to entities documented in UniProtKB entries. PRO relates to the GO in that PRO’s representations of organism-specific protein complexes are subclasses of the organism-agnostic protein complex terms in the GO Cellular Component Ontology. The past few years have seen growth and changes to the PRO, as well as new points of access to the data and new applications of PRO in immunology and proteomics. Delo se loteva vprašanja aktualnosti pojma subjekta v filozofiji in politiki skozi analizo tistih avtorjev, ki so ga domnevno najbolj radikalno pokopali: pokaže, da vznik radikalno novega in problematika subjekta zavzema osrednje mesto v Deleuzovi filozofiji, kakor tudi v Lyotardovi pozni misli posvečeni praznini, v Foucaultovi obravnavi biopolitike in biooblasti, v delu Negrija in Hardta o Imperiju, ter nazadnje v Badioujevi predelavi temeljnih filozofskih kategorij biti, resnice in subjekta, na osnovi katerih je dandanes znova možna renesansa filozofije. Ta ima po (...) Badiouju štiri razsežnosti, ki se v delu Vznik subjekta nenehno prepletajo: »V osnovi želja filozofije implicira razsežnost upora: ni filozofije brez nezadovoljstva mišljenja v njegovem soočenju s svetom, kakršen je. Pa vendar želja filozofije vključuje tudi logiko: to je, prepričanje v moč argumenta in uma.«. (shrink)

Neuroenhancement involves the use of neurotechnologies to improve cognitive, affective or behavioural functioning, where these are not judged to be clinically impaired. Questions about enhancement have become one of the key topics of neuroethics over the past decade. The current study draws on in-depth public engagement activities in ten European countries giving a bottom-up perspective on the ethics and desirability of enhancement. This informed the design of an online contrastive vignette experiment that was administered to representative samples of 1000 respondents (...) in the ten countries and the United States. The experiment investigated how the gender of the protagonist, his or her level of performance, the efficacy of the enhancer and the mode of enhancement affected support for neuroenhancement in both educational and employment contexts. Of these, higher efficacy and lower performance were found to increase willingness to support enhancement. A series of commonly articulated claims about the individual and societal dimensions of neuroenhancement were derived from the public engagement activities. Underlying these claims, multivariate analysis identified two social values. The Societal/Protective highlights counter normative consequences and opposes the use enhancers. The Individual/Proactionary highlights opportunities and supports use. For most respondents these values are not mutually exclusive. This suggests that for many neuroenhancement is viewed simultaneously as a source of both promise and concern. (shrink)

Von Peter Sloterdijk kann man zu Recht sagen, daß jeder seiner Aufsätze, jeder seiner Vorträge auch ein ungeschriebenes Buch ist.

Was in Theorien an Auflösungsvermögen verkraftet werden kann und muß, findet in der Welt, die durch Theorie rekonstruiert werden soll, eine Parallele: Was der Mensch sei, ist nicht einmal menschen- und lebensweltlich eine klare Kante. In den Humankatastrophen der letzten hundert Jahre wird er zu einer verfeuerungsfähigen Biomasse. Man kann kaum den Eindruck gewinnen, daß sich daran etwas wirklich geändert hat. Es wird Tag für Tag hekatombenweise gestorben, gemordet, gefoltert. Der Mensch wird definiert als das Wesen, das man töten kann (...) durch Hunger, Krankheit, Folter, Mord und Krieg, namenlos, zahllos, heute hier, morgen dort. Man könnte sich denken, daß die Lehre vom Menschen, die Anthropologie, ihren Grund verliert und eigentlich zur Thanatologie konvertieren müßte. Und dank Gentechnologie erscheint am Horizont die Vision, der Mensch sei nicht mehr jemand, der nach dem Bilde eines Gottes geformt sei, sondern nunmehr jemand, der in den Stand versetzt wurde, sich ein machbares Bild von sich selber zu schaffen, die Exemplare der humanen Population so zu formatieren, daß sie dem alten Ideal der Kalokagathia entsprechen: tugendhaft, schön, alters- und behinderungslos, krankheits-, rausch- und drogenfrei, angemessenen Verstandes eine Idee der Devianzausmerzung, könnte man sagen, die Idee einer leid- und schicksalsfreien Ruhewelt, einer Re-Animation des Paradiesgedankens ohne Adam und ohne Eva. Das Verhaltnis zwischen Mensch und Natur spielt in vielen moralischen und politischen Fragen eine wichtige Rolle. Es ist philosophisch nicht als Verhaltnis zweier vorgegebener Gegenstande, sondern als Gegensatz von naturalistischer und kulturalistischer Perspektive der Wissenschaften auf den Menschen zu fassen. Dabei geht es um eine Auflosung der Konfrontation, die sich aus Alleinvertretungsanspruchen der beiden Perspektiven gegenuber der jeweils anderen ergibt, in eine begrundete Arbeitsteilung natur- und kulturwissenschaftlicher Beitrage. Am Beispiel von Poiesis als handwerklicher Herstellung und zweckrationaler Technik lasst sich mit (...) den Mitteln der Handlungstheorie ein anthropologisches Spezifikum bestimmen, das die Geschichtlichkeit des Menschen und seiner Kultur notwendig bedingt. Dabei erweist sich technische Zweckrationalitat als gemeinschafts- und geschichtsabhangig; sie bildet einen erkenntnistheoretisch und ethisch erheblichen Aspekt des Menschen. (shrink)

Die vorliegende Untersuchung setzt sich mit jenen Werken auseinander, die im frühen 18. Jahrhundert hermeneutische Problemgehalte ansprechen. Gleichsam als Vorbotin einer Kritik der Verstandeserkenntnis wird in der rationalistischen Aufklärung eine Bewegung spürbar, die geschichtshermeneutische, erkenntnis- und moralkritische Ansätze hervorbringt und in Opposition zu allgegenwärtigen nomologischen Vorstellungen die Frage nach dem ganzen Menschen neu formuliert. Diese ästhetisch-hermeneutische Kritik bezieht sich nicht nur auf die Gesetzeswissenschaften, sondern auch auf geschichtliche Problemstellungen, so daß die hier thematisierten Diskussionen über Gemeinsinn, empfindsame Moralkritik und ästhetische (...) Im Vergleich mit anderen rechtsphilosophischen Schriften Platons - etwa der Politeia - hat der Dialog "Kriton" in Kontinentaleuropa bisher wenig Beachtung gefunden. Anders als im anglo-amerikanischen Wissenschaftsraum fanden sich bisher nur vereinzelte Stellungnahmen in Gesamtdarstellungen zu Platons Philosophie.Die Monographie folgt den Ereignissen und Gedankengangen der Dialogpartner und rekonstruiert erstmals umfassend den rechtsphilosophischen Gehalt der Schrift, das heiat die zugunsten der Pflicht zum Rechtsgehorsam vorgebrachten Argumente. Diese Rekonstruktion fuhrt zu der Erkenntnis, daa Platon im "Kriton" eine konsistente und mit seinen anderen (...) (Fruh-) Schriften kompatible Begrundung fur die Pflicht zum Rechtsgehorsam geliefert hat, die im Kern auch heute noch bedenkenswert ist.Die Analyse ist daher nicht nur fur eine vollstandige Erfassung der gewichtigen Rechtsphilosophie Platons, sondern auch fur die gegenwartige Diskussion um die Gehorsamspflicht von Nutzen. Review of Ernst, "Das Problem des Wissens".

Human dignity seems very important to us. At the same time, the concept ‘human dignity’ is extraordinarily elusive. A good way to approach the questions “What is it?” and “Why is it important?” is to raise another question first: In virtue of what do human beings have human dignity? Speciesism – the idea that human beings have a particular dignity because they are humans – does not seem very convincing. A better answer says that human beings have dignity because and (...) insofar as they are persons. I discuss several versions of this idea as well as several objections against it. The most promising line of analysis says that human beings cannot survive psychologically without a very basic form a recognition and respect by others. The idea that humans have a special dignity is the idea that they owe each other this kind of respect. All this also suggests that human dignity is inherently social. Non-social beings do not have dignity – nor do they lack it. It is because we are social animals of a certain kind that we have dignity – not so much because we are rational animals. (shrink)

Im Jahre 1947 erscheint eine Aufsatzsammlung Alfred North Whiteheads unter dem Titel Essays in Science and Philosophy . Unter der Rubrik ,,Personal“ finden sich dort Whiteheads mit An Appeal to Sanity betitelte Überlegungen, die er im März 1939 am Vorabend des 2. Weltkriegs erstmals veröffentlicht hatte, und die er nach dem Ende und der Erfahrung dieses Krieges 1946 um einige Bemerkungen erweiterte. Die zentrale Feststellung, mit der Whitehead das sieben Jahre zuvor Beschworene gleichsam aus den Angeln hebt, bezieht sich auf (...) die Sorge, es möge für die Zukunft der Zivilisation keine Hoffnung geben, es sei denn innerhalb einer von der UNO angeregten und von ihr durchgesetzten Charta ethischer Prinzipien. – Die vorliegende Studie nimmt diese Erweiterung und substantielle Veränderung Whiteheads ursprünglicher Gedanken von 1939 zum Anlass und geht der Frage nach, ob Whitehead je nach historischer Lage einmal so und dann wieder anders argumentiert, oder ob sich in der Behandlung des Themas ,Krieg' metaphysische Grundüberzeugungen unverändert und doch in einer je anderen historischen Gestalt zeigen. The article presents Observations of Catholic moral teaching from the perspective of Protestant theology in the form of theses. In a first section, main traits and distinctions, especially in the tradition of the claimed Magisterium of the Catholic Church, are outlined. The second section deals with typical Protestant critiques and misunderstandings of the Catholic approach. Following this article they are caused by confounding different Ievels of argumentation: the Ievels of a) the doctrines of teaching, b) the acting of teaching and (...) c) the comprehensive cultural practice. Unfolding these distinctions and their particular functionings the article draws the surprising conclusion that Protestant theology and ethics often do not match Catholic thinking although they think they do by using the same words. This conclusion may have a huge impact on ecumenical ventures and the Protestant self-understanding. A third section, a brief outlook on these questions, completes the train of thoughts. (shrink)

The present article explores the historical sections of Grotius's De iure praedae Commentarius bearing the following fundamental but very important questions in mind: What did Grotius actually know about the Portuguese Estado da Índia at the time of drafting the treatise between 1604 and 1606/8? What did he know about the Luso-Asian trading regime or Asian trading practices at large? Using the published correspondence of Grotius, archival documentation, manuscript fragments as well as unpublished reading notes and drafts, a case will (...) be made that he had in fact few sources at his fingertips. This insight serves fundamentally to refute older assumptions, expounded notably by Robert Fruin and C.H. Alexandrowicz, that the young Grotius conducted independent research in the VOC company archives, or had immersed himself with Asian maritime codes or commercial practices. (shrink)

Die Geometrie ist seit Euklids Elementen nicht nur Vorbild fur Theorieform und Wissenschaftlichkeit. Sie hat auch seit der Antike die erkenntnistheoretische Diskussion uber das Verhaltnis von Wirklichkeit und Erkenntnis beeinflusst. In ihrer formalaxiomatischen Auffassung der physikalischen Geometrie durch A. Einstein wird die Wissenschaftstheorie des 20. Jahrhunderts in ihren Mehrheitspositionen auf einen Logischen Empirismus festgelegt. Vollstandig ausgeklammert bleibt dabei das philosophische Problem der Gegenstandskonstitution. Wovon ist Geometrie eine Wissenschaft, und wodurch erhalt sie ihre Passung auf die Anwendungen in Handwerk, Technik und (...) messender Naturwissenschaft? (shrink)

Aufgrund sorgfältiger Untersuchung in phänomenologischer Erkenntnishaltung können die Struktur des Lysis, die künstlerisch-ideelle Gedankenbewegung und damit die Einheit von philosophisch-künstlerischer Gestalt und philosophischem Gehalt des Dialoges in einer strengeren Weise erfasst werden als bisher. Dadurch werden ganz neue Beziehungen zu Gestalt und Gehalt der Politeia sichtbar. Sie erlauben im Verein mit grundsätzlichen Überlegungen zur Methode der datierenden Sprachstatistik und weiteren Beobachtungen, den Lysis chronologisch nach der Politeia anzusetzen - in der Trilogie Phaidros - Lysis - Euthydemos. So können die interpretatorischen (...) Einschränkungen durch das Vorurteil, der Lysis sei ein Frühdialog, überwunden werden. Die positive Interpretation des Dialoges und ihre Vertiefung durch die platonische Prinzipienlehre werden damit auf ungewohnte Weise stärker begründet. (shrink)

Der Essay geht der Frage nach, ob es eine besondere, methodisch einheitliche Art der Philosophie gibt, die man mit gutem Grund ‚Analytische Philosophie’ nennen könnte. Als Beispiele werden Kriterien wie Klarheit, gedankliche Übersicht und das Gewicht von Argumenten besprochen. Das Ergebnis der Suche ist negativ. Die These lautet, dass die interessante analytische Philosophie nicht deshalb interessant ist, weil sie analytisch ist, sondern weil sie gute Philosophie ist.
